Demonstration of a humoral inhibitor of the Na+-K+ pump in some models of experimental hypertension.

چکیده
We have previously shown that ouabaln-sensitive Rb uptake, a measure of Na-K pump activity, is decreased in the blood vessels of dogs with one-kidney, one wrapped hypertension and rats with onekidney, DOCA, saline hypertension. We here extend the study to rats with one-kidney, one clip and reduced renal mass-saline hypertension. We also assayed supernates of boiled plasma from three of these models for ouabain-sensitive Rb uptake suppressing activity. Finally, we examined the influence of the anteroventral third ventricle (AV3V) lesion in the rat on vascular pump and plasma supernate activities. We found that ouabain-sensitive Rb uptake is suppressed in the tail arteries of rats with one-kidney, one clip and reduced renal mass hypertension and that plasma supernates from these rats and from dogs with one-kidney, one wrapped hypertension suppress Rb uptake when applied to tail arteries from normal rats. We also found that, in the volume-expanded state, rats with AV3V lesions had higher vascular ouabain-sensitive Rb uptake than rats with sham lesions and evidence for decreased inhibitory activity of the plasma. These findings suggest that reduced vascular Na-K pump activity is common to several models of experimental hypertension and that this defect results from a heat-stable ouabain-like agent in plasma that originates in or is influenced by the AV3V area of the brain. Humoral sodium transport inhibitor in acute volume expansion and low renin hypertension.
This review summarizes our bioassay methods for determining the level of humoral sodium pump inhibiting factor after acute volume expansion in experimental animals and humans, and in low renin experimental and human essential hypertension. In brief, ouabain-sensitive 86Rb uptake and membrane potential in blood vessels from normal animals are measured after incubation in plasma supernate from ex...
متن کاملSodium-potassium pump activity in reduced renal-mass hypertension.
We have previously reported that ouabain-sensitive "Rb uptake, a measure of Na-K pump activity, is decreased in blood vessels of animals with several low renin, and probably volumedependent models of hypertension likely due to the action of a circulating sodium transport inhibitor. In this paper we summarize a detailed study of these and other related parameters in a model known to be volume ex...
متن کاملAbnormalities of membrane transport in hypertension.
In this review, postulated passive and active fluxes of sodium, potassium, and calcium across the sarcolemma of the normal vascular smooth muscle cell are first summarized. Some practical problems encountered in their measurement are also mentioned. The review then considers how these fluxes appear to be altered in various forms of hypertension in animals and humans. Emphasis is given to abnorm...
متن کاملO-13: Na+/K+-ATPase Alpha1 Isoform Mediates Ouabain-Induced Expression of Cyclin D1 and Proliferation of Rat Sertoli Cells
Background: Novel roles for the interaction of cardiotonic steroids to Na+/K+-ATPase have been established in recent years. The aim of the present study was to investigate the intracellular signaling events downstream the action of ouabain on Na+/K+-ATPase in Sertoli cell obtained from immature rats. Treatment of Sertoli cells with ouabain (1 μM) induced a rapid and transient increase in the ex...
متن کاملThe vascular Na+-K+ pump in experimental hypertension.
We assessed the role of putative circulating ouabainlike factor(s) on in vivo arteriolar function in rats with very early (less than 7 days; mean, 3 days) and chronic (greater than 4 weeks) benign, one-kidney, one clip (1K1C) hypertension. Thus, we measured vascular responses in vasodilated (nitroprusside or adenosine), vascularly isolated, innervated hindlimb vascular beds of chloralose-anesth...
